Incretins are gut-derived hormones that stimulate insulin secretion from pancreatic beta cells in response to nutrient intake. The two principal incretins are GLP-1 and GIP, both released after meals. The "incretin effect" — that oral glucose stimulates more insulin release than IV glucose at the same plasma level — is diminished in type 2 diabetes, explaining why incretin-mimetic drugs work.
